主要研究方向

(1) 高通量免疫组库测序

免疫组库测序是通过检测T/B 细胞受体互补决定区(CDR 区)来全面评估免疫系统的多样性,深入挖掘免疫组库与健康或疾病的关系。免疫组测序技术是目前免疫系统研究的最前沿、最热点技术,广泛应用在疫苗开发、肿瘤免疫治疗、自免疫疾病、药物研发等等医学研究的各个领域。

我们课题组拥有世界领先的免疫组库测序与分析技术,能够实现了对TCR/BCR的七条链无偏差、定量检测。以此基础技术上,我们和多个国内顶尖医学团队建立了牢固、深入的合作的关系,包括呼吸疾病国家重点实验室(钟南山院士、陈凌教授)、湘雅二院(陆前进教授、吴海竞教授)、上海质子重离子医院(蒋国良教授)、江苏省人民医院(杨涛教授)等,合作发表高水平研究论文多篇、申请科研项目多项。

(2) 分子诊断技术研究

分子诊断技术是指以DNARNA为诊断材料,用分子生物学技术通过检测特定核酸靶标的存在、缺陷或表达异常,从而对人体状态和疾病作出诊断的技术,其研究对疾病的预防、预测、诊断、治疗和预后具有重要意义。

课题组在国内最早开展基于纳米技术的高通量、自动化核酸检测方法的研究。多年来,我们结合微阵列技术、磁性纳米颗粒、荧光杂交技术以及单碱基延伸技术发展了多种高通量自动化核酸检测方法及其相关技术,并取得了一系列重要的研究成果。我们发展的基于磁性纳米颗粒的SNP检测技术被作为标准方法被编入行业工具书,同时获得国际同行的高度评价。相关研究先后获得国家973、自然科学基金及其他省部级重点项目支持。

(3) 全自动一体化POCT系统

基于核酸检测的全自动一体化POCT系统凭借其灵敏度和准确度高、携带方便、操作简单、检测时间少、对环境无要求及成本低的优点,已经成为体外诊断行业发展的主流。尤其是在后疫情时代,医疗市场对于此类系统的需求非常迫切。

本人在美国Hudsonalpha生物技术中心和iCubate公司工作器件,作为核心团队成员发展了新一代“全自动多重分子诊断系统”——“iCubate”系统及其配套试剂盒并且获得FDA认证。课题组还与上市公司万孚生物合作完成了该系统的系统升级与国产化,并且取得了三类医疗器械认证。目前课题组开展以一体化检测器件为载体的小型全自动分子诊断系统研究,重点利用光学、电学、磁学检测技术对DNA、蛋白质等生物大分子进行检测。该方面研究受到国家重点研发计划,自然科学基金和湖南省重点专项等资金的支持。
